Protocol Overview

Total RNA from liver and small intestine was isolated using TRI Reagent® as per the manufacturer’s instructions. In general, 1 mL of TRI Reagent® was added to 50-100 mg of tissue (approx) and homogenized in glass homogenizer. The homogenate was further processed for phase separation and centrifuged. To the clear aqueous phase, isopropanol was added to precipitate the RNA. The contents were vortexed and centrifuged. RNA pellet was washed with 75% ethanol, air dried and dissolved in nuclease free water. The RNA samples were stored at -20°C.

Results Summary

The purity of RNA obtained was good and A260/A280 ratio was between 1.6-1.9. The integrity of RNA was also good.

TRI Reagent® should be stored at 4°C
