The aim of our study was to transfect siRNA in mammalian cells to achieve siRNA knockdown at the protein level. Selection of HiPerFect Transfection Reagent from Qiagen was based on merits like a rapid transfection method and low cytotoxicity after transfection.
Mammalian cells were cultured and siRNA at 200 molal concentration was transfected using HiPerFect transfection reagent. All steps of transfection were followed as per manufacturer's instructions. Transfected cells were incubated for 48 hrs to deplete siRNA specific protein levels. After transfection, proteins were extracted, resolved in 10% SDS-PAGE, western trnasferred and probed with specific antibodies. ECL detection was used to visualize HRP activity.

The transfection protocol was rapid and no or very little cytotoxicity was observed. Transfection efficiency was very high as almost 90% of siRNA specific protein was depleted.
