I expressed our gene of interest (ESAT6) in a bacterial system by employing standard transformation. I screened out colonies using a qPCR assay wherein I used RT2 SYBR green qPCR mastermix to quantify the mRNA copy number. Overall, I am highly satisfied with this reagent.

cDNA from transformed colonies
I used the standard manufacturer's protocol to amplify the cDNA prepared from the transformants. I ran 20 cycles and used Applied Bioscience's Real Time PCR instrument to perform the PCR.

The positive clones clearly showed marked increase in the expression of ESAT6.
