I study immune response in asthma and use cell proliferation assays quite often. I use this anti-BrdU staining kit to study proliferation of immune cells in response of various stimuli and it always gives me satisfactory results.
Cell proliferation assay
Cells
Cells were cultured in the presence of stimuli and BrdU for 72 hrs at 37 degrees, cells were harvested and stained for surface markers then fixed using 1X BrdU staining buffer for 30 minutes, followed by DNase I treat for 37 degrees for 1 hour. After washing, cells were stained with anti-BrdU FITC conjugated antibody and analyzed thru FACS after washing twice.
Fix/Perm cell no more than 30 minutes at room temperature
Kit generates neat data for cell proliferation and detects cells in actively dividing state.
