Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Human PBMCs

Primary Incubation

10 mins room temperature (Fc Receptor blocking, 5µl for a 100µl cell suspension containing 2 milllion cells)

Blocking Agent

TruStain FcX blocking solution (BioLegend)

Secondary Incubation

30 mins on ice in the dark, 2µl of supplied antibody solution

Tertiary Incubation

None

Detection

Flow Cytometry

Results Summary

Reliable labeling of CD16 positive cells in PBMCs, also good antibody for labeling subpopulations in a monocyte-enriched cell fraction. Really bright fluorescence (picture), can be used with CD14 FITC to classify subpopulations of monocytes.

Additional Notes

Fc-block with TruStain Fc Receptor blocking solution, specifically designed to analyze CD16 (Fc Receptor) after blocking it.

Summary

The Good

Reliable, easy staining with quite quick incubation time

The Bad

Blocking needs to be performed, sometimes with a rather big excess of the blocking solution, 3 washes

The Bottom Line

Very good reliable antibody for labeling CD16 positive cells in PBMC and monocytotic samples. Bright fluorescence, easy to handle.
