Company:

Thermo Fisher Scientific (Invitrogen)

Product Name:

Prolong gold antifade reagent with DAPI

Catalog Number:

P36931

This is a mounting solution for microscopy cell viewing. First of all, this mounting solution has DAPI in it, which brings up the price as compared to a regular mounting solution. Since it already has DAPI inside, you cannot adjust the concentration of DAPI, so you have to adjust fluorescent exposure time to get a good figure. DAPI itself is a very specific DNA dye, so no problem with dye specificity. However, if you have multiple types of cells to work with, and you need an equal working concentration of DAPI, this is not the one to go with because the only way to have an equal DAPI concentration is to add same amount of mounting solution to the coverslips while sealing. You cannot wash the coverslips anymore after applying mounting solution.

Experimental Design and Results Summary

Application

Fluorescent microscopy

Starting Material

Bacteria whole cells

Protocol Overview

Bacteria were fixed and permeabilized, then spun onto coverslips. This mounting solution was then applied.

Tips

Thaw before use, takes a pretty long time to thaw; use a small amount (~5ul)

Results Summary

After several trial times, I got the image I wanted. It is a fluorescent picture of DAPI, quite clean with no background. The working concentration of DAPI is cell-dependent, so if you do not know the concentration to use, buy a separate dye to try.

Summary

The Good

It works for my purposes, no need to have a separate dye.

The Bad

Takes time to thaw, hard to optimize concentration

The Bottom Line

Cell-dependent mounting solution, choose with caution
