I use this staining kit to measure T-cell proliferation induced by our stimuli. Since I have FITC conjugated antibodies in my staining cocktail, I use this staining kit to avoid compensation issues.
Flow cytometry
Human and mouse CD4+ T cells (Any other cells also)
I used this to stain 1 million cells with 1 microliter of 5 micro mole working solution stain for 20 min at 37c dark. Then wash with 1% FCS containing PBS for 5 min.
Avoid adding excessive or inadequate stain.
I used to get two distinct populations of dividing and non-dividing cells upon dilution of this dye in proliferating cells during cell division.
