Measuring protein concentration is one of the first steps in our workflow. We use this assay because its 96 well microplate format works well with our typical concentration range in cell lysates (a few ug per ul) and we see no interference with our buffers.

Protocol Overview

The procedure is straightforward and the manual is easy to understand. Working reagent is made by combining two components together. The working reagent plus either diluted protein standards or samples are loaded into a microplate, shaken for 30 seconds, then incubated for 30 minutes at 37C before reading at ~562 nm.

Tips

Quadratic fit works better than linear fit because the absorption begins to plateau

Results Summary

Scraping a 100 mm dish into 100 uL of lysis buffer, we dilute the resulting post-nuclear homogenate 3 or 4 fold to be within range. We measure 300-500 ug of total protein per dish depending on cell density.
