I perform a lot of plasmid minipreps daily. I prepare these for many different purposes, including restriction analysis, cloning, sub-cloning and DNA sequencing. I have been using the spin miniprep kit for a long time and I have found it very efficient in my routine. I chose Qiagen's miniprep kit for the sake of availability at first and found it very useful in my hands with excellent results. I get pure supercoiled DNA without any contamination. This has been my kit of choice for small size plasmid minipreps for years.

Starting Material

E. coli overnight culture

Protocol Overview

Centrifuge 5 ml of overnight E. coli culture. Spin to collect the cells. Mix the pellet in 250 ul of resuspension buffer. Add 250 ul of lysis buffer. Gently mix, then add 350 ul of neutralization buffer. Spin and collect the supernatant in the spin column. Spin the column for 1 min and wash with 250 ul of washing buffer. Spin the column without any buffer to let partially dry. Elute the DNA with 50 ul of elution buffer.

Tips

Always wash the spin column with washing buffer to get purest form of DNA.

Results Summary

In my routine, I use washing buffer always. This gives me purest supercoiled DNA. It is said in the manual that it is optional but I highly recommend doing this. The DNA obtained by this kit is high quality and can be used for a lot of downstream applications.

Summary

The Good

Easier to follow, gives pure plasmid DNA.

The Bad

A bit more expensive than others, the amount of DNA is pure but a bit lower in quantity.

The Bottom Line

In conclusion, I would highly recommend using Qiagen's plasmid DNA miniprep kit. This is a very efficient protocol for plasmid isolation and takes around 30 minutes at maximum.
