We have had difficulties getting a good signal on our Licor digital Western blot imager using standard ECL substrates. Bio-Rad's Clarity reagent solved this problem with a good signal after a shorter imaging time.

PVDF membranes with cytoplasmic lysates

Protocol Overview

750 ul each of Clarity reagents A and B were mixed in an eppendorf tube and applied to a PVDF membrane containing cytoplasmic lysates (20ug loaded/lane). The lysates were from human B cell lines that were either untreated or stimulated overnight with monosodium ureate crystals. After electrophoresis in 4-12% of a Bio-Rad TGX gel, lysates were transferred via standard protocols. Membranes were probed with rabbit anti-human IL-6 polyclonal antisera, followed by a goat anti-rabbit secondary-HRP. Blots were imaged using a Licor C-Digit blot scanner for six minutes.

Results Summary

Images from the experiment were clear with crisp bands and no background. The reagent solved a problem that had been plaguing several labs using standard ECL HRP substrates.
