Extracting RNA from cell lines or from tissue specimens has the historic connotation of being an arduous process. This kit, from Qiagen, makes RNA extraction easy with pure RNA content after extraction. The reagents are inclusive within the kit and it takes roughly 30 minutes to extract RNA from cell lines with repeatable purity values.

Protocol Overview

Protocol was followed verbatim: Initially adherent cells were trypsinized and pelleted. The cell pellet was resuspended in fresh media. The specimen was then re-centrifuged and the protocol was followed thereafter.

Tips

Requires absolute ethanol. Would spin a minute longer to dry the membrane towards the end of the protocol.

Results Summary

RNA analysis demonstrated a pure specimen. The RNA extracted was utilized for RT-PCR/qPCR analysis of gene expression of glioblastoma cells.
