Best Result from Isolectin GS-IB4, Alexa Fluor 488 Conjugate

Protocol Overview

Mouse eyes were enucleated and fixed in 4% paraformaldehyde for 1Hr at RT. Intact retina were isolated under a microscope and permeabalized overnight by incubating in 0.5% Triton X-100 at 4 °C. Permeabalized retina were washed with PBS and incubated overnight at 4 °C with isolectinB4 (10 μg per ml) containing 1 mM CaCl2, 2 mM NaN3 and 1× PBS. Stained retina were flat mounted and observed under epifluorescence microscope. Green colour from microscope software was selected for image capture.
