We followed the manufacturer protocol. In brief, 3 volumes of the ERC is added to 1 volume of sample and loaded on to the qia quick column and spun. The column is washed with 750 microliters of PE buffer and eluted with 50 microliters of elution buffer
The ERC buffer colour should be yellowish or else add 10 microliter of 3M sodium acetate to regain its PH
Good quality of DNA is obtained with a moderate recovery.
The DNA fragment should be at minimum 75 base pairs for purification
High quailty DNA is obtained with low salts
Cannot use for amplicon of less than 75 base pairs
Go for this kit when you want to purify DNA amplicon greater than 100bp
