DC140

I am looking at whether yogurt consumption could improve intestinal barrier function in an obese population. Compromised barrier function could lead to increased exposure to endotoxins, such as LPS. CD14 is one mediator involved in the LPS-TLR4 pathway. Therefore, it can serve as an indicator of endotoxin level, which in turn reflects the barrier function.

Experimental Design and Results Summary

Application

For sCD14 measurement in human plasma

Starting Material

Human plasma

Protocol Overview

1. Prepare wash buffer and standard solution. 2. Add assay diluent. 3. Add standard and diluted samples, and incubate. 4. Wash the plate. 5. Add conjugate and incubate. 6. Wash the plate. 7. Add substrate solution (mixed the color reagent within 15 min of use), ad incubate. 8. Add stop solution. 9. Read

Tips

Figure out the appropriate dilution factor for your sample. Remember to vortex to mix well when diluting samples.

Results Summary

Make standard curve with 4 parameters. Quantify sample solution based on the standard curve. Perform back calculations to find the original concentration after accounting for dilution factors.
