I used this IL-13 to activate JAK/STAT signaling in my cells. IL-13 increased STAT6 phosphorylation, which was my assay for activation of JAK/STAT signaling.
Activation of signaling pathways
Primary Human Epithelial Cells
Incubated primary epithelial cells for 1 hour with or without 3 ng/ml IL-13. Made whole cell lysates and then ran a Western blot probing for phosphorylated STAT6 (pSTAT6), which has been shown to increase following IL-13.
Dissolve IL-13 in 0.1% BSA solution
The IL-13 dramatically increased pSTAT6 levels compared to non-IL-13 treated lysates. This indicates that IL-13 is being recognized by the IL-13 receptor on the epithelial cells and is causing the expected increase in pSTAT6.
