I am working on an intervention study to look at whether yogurt consumption can reduce chronic inflammation, which is linked to the development of various diseases. Il-6 has been used as biomarker for inflammatory levels in numerous reports.

Heathy human EDTA plasma

Protocol Overview

1) Bring samples and kit to room temperature. 2) Prepare the standard stock solution. It takes a minimum of 15 mins for the standard to dissolve; so plan ahead for that. Be advised: for healthy human plasma, there is no need to dilute the plasma samples. 3) Add dilution buffer to the plate (do not forget this step!) 4) Follow the kit insert: load samples, control and standard-----incubate on orbital shaker---wash---add conjugate--incubate on orbital shaker--wash----add substrate---add amplifer---add stop solution--read (takes about 6-7 hours due to the long incubation period)

Tips

Prepare standard stock solution, substrate solution and amplifier solutions ahead.

Results Summary

The kit works pretty well. I got acceptable cvs within and between batches.
