This antibody was used in a study that used VEGFR2 as a marker for endothelial cell (EC) differentiation from MSCs. Several antibodies and concentrations were tried, but ultimately Cell Signaling's #2472 gave the best results for IF visualization of VEGFR2.

Primary Incubation

1:200 in blocking solution overnight at 4°C

Blocking Agent

5% BSA + 0.3% Triton X-100 in 1x PBS

Secondary Incubation

5 ug/mL Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor® 594 conjugate (Life Technologies cat #A-11037) in blocking solution for 1 hour at room temperature

Tertiary Incubation

Endothelial cells also stained with DAPI (1:5000 of 5 mg/mL stock in 1x PBS for 3 minutes at room temperature) for nuclear localization

Detection

Nikon Eclipse Ti Microscope with fluorescent filters, 20x objective

Results Summary

Cell Signaling's VEGFR2 antibody worked well for immunofluorescence antibodies when paired with an AlexaFluor-conjugated secondary. Minimal background and non-specific binding was observed. Specific for cells expressing VEGFR2 when compared to non-differentiated MSCs and a non-VEGFR2 expressing control.
