The focus of my project is elucidation of the physiological role of endocytic regulatory protein EHD4 in the kidney. EHD4 is expressed in the inner medullary collecting duct (IMCD) of the renal nephron. IF staining with a cell specific marker showed that EHD4 is present in the intercalated cells of IMCD. Intercalated cells are responsible for regulating urine pH. Urine samples from wild type and EHD4 knock-out mice were collected for 24 hours. However, the sample sizes were too small to be measured by regular pH meter. So I used this micro pH meter to serve the purpose.

The glass electrode was connected to a electric pH meter using the BNC connector. The glass electrode was filled with reference filling solution and calibrated immersing the glass electrode in standard solutions (pH 4,7 and 10). After calibration the electrode was immersed in sample solutions and readings were taken.
Be careful with glass electrode. It should be handled with care. For best calibration and results, the samples should be in the same temperature as the buffers.
The readings generated the pH of the urine samples. I took two readings per samples and the readings were consistent. Try not to move your samples too much; slight movement can disturb readings.
