The aim of my research was to measure NPY concentration in plasma from cows exposed to different nutrition during gestation. I used the product to measure NPY in plasma from cows. I followed the protocol and used the equipment recommended. However, I was not able to obtain consistent results within animals or a consistent standard curve running duplicates. I selected the product because it was a bovine specific kit.

Protocol was easy to follow. However, no results were obtained since readings were too inconsistent.
ALWAYS run your standard curve in duplicates
No results obtained. In the figure below, the x axis is the log of the concentration that was specified on the protocol for the standards that came with the kit. The y axis is the log of the actual concentrations I got running the standards in the assay. The biggest problem I had is first I could not validate the assay since the standards never ran in the concentration specified on the kit. Second, whenever I ran duplicates the intra-assay was higher the 30, which would be unacceptable for a publication.
