Our experiments assess the tendencies of environmental pollutants to affect dendritic cells' protein expression. We incubated Bone-Marrow Derived DCs with pollutant and collected data via flow cytometry. CD86 is a costimulatory molecule that we expect to increase in expression after incubation with the pollutant. Biolegend's BV510 is slightly difficult to optimize, which is partially due to CD86 often showing a shoulder population.

Experimental Design and Results Summary

Flow Cytometry

Sample

Bone marrow derived dendritic cells

Primary Incubation

2uL of 50ug/mL per million cells for 30 minutes at 4 celsius

Results Summary

Brilliant Violet 510 enables more sensitive assessment of CD86 expression than other fluorophores. It is difficult to optimize & uses more antibody than the Biolegend recommends per million cells.

The Bottom Line

I would recommend this product, but titrate it with your own samples before using it in a real assay. The protein amount needed will likely be different than what is recommended.
