Protein SUMOylation is an important post-translational modification required for normal physical functions of a cell. Ubc9 is an evolutionarily conserved SUMO E2 enzyme involving in the SUMO conjugation to its substrate proteins to regulate cell cycle progression, p53 signaling pathway, and DNA damage response.

Experimental Design and Results Summary

Applications

Western Blot

Sample

HeLa human cancer cell line

Primary Incubation

Dilution 1:1000 in 5% non-fat milk/TBS-Tween20. Two to four-hour incubation at room temperature.

Blocking Agent

5% non-fat milk/TBS-Tween20

Secondary Incubation

HRP-conjugated anti-rabbit IgG

Tertiary Incubation

NA

Detection

HyBlot ES autoradiography film

Results Summary

HeLa cells were transfected with control and different Ubc9 siRNAs for 66 hours followed by the direct lysis of samples in 2x sample buffer. The Western blot result showed that Ubc9 was detected in control siRNA treated, but not the Ubc9 siRNA treated samples. Tubulin was used as loading control.
