Phosphorylation of Chk1 at S317 is a hallmark of the activation of cell cycle checkpoint. The goal of this experiment was to examine whether, after UV irradiation, Phospho-Chk1 (S317) can be detected in HeLa cells by immunofluorescence.

Dilution 1:50 in PBS/0.05% Tween20/3% BSA blocking buffer. Overnight incubation at 4 C.
3% BSA / 0.05% Tween20 in PBS
Cy3-conjugated anti-Rabbit, second antibody at dilution of 1:250 and 1 hour incubation at room temperature

Fluorescent microscope
HeLa cells irradiated by UV or left undamaged were fixed by methanol and compared by immunofluorescence using Phospho-Chk1(S317). The UV treated samples showed significant increase of Chk1pS317 compared to undamaged cells.
