The stimulation of T cells is carried out by different methods. In order to check if your stimulation worked or not, there are many factors you can look at. CD69 is one of these markers. Lindsey WB et al (Cytotherapy. 2007;9(2):123-32) show that it's a very good marker.I use this antibody because it's a good antibody, easy to use and relatively cheap.

Results Summary

In the example flow plot shown, T cells were either incubated with media only (red histogram) or stimulated with PMA/ionomycin using a standard protocol. CD69 can be seen strongly positive on the PMA-stimulated cells (blue histogram).
