An easy to way to measure cell death in a plate-based assay. This kit allows the scientist to rapidly measure the levels of caspase-3/7 activation in vitro. This assay complements other commonly used assays for cell death such as Annexin V/PI staining.

Protocol Overview

MEFs were seeded in 96-well plates and stimulated. The luciferase-based caspase assay was used to measure Caspases-3/7 activity following stimulation according to manufacturer’s protocol. Samples were read on a multilabel automated plate reader (Perkin Elmer) and triplicates were performed for each condition (means ± SD are presented).

Tips

Reduce the volume by 50% to extend the number of assays that can be performed.

Results Summary

WT (black) and KO (white) MEFS were stimulated as shown and caspase-3/7 activity was measured. Results are presented as ''fold activation'' compared to the control for each genotype (ST, Staurosporine).
