Our research focuses on microRNAs, especially in how they can be used in medical applications.

I have used the Life technologies Exosome Isolation reagent to isolate exosomes from human serum. These were then used to extract RNA and quantitate microRNA expression by qRT-PCR.

250ul aliquots of human serum samples

Protocol Overview

The protocol is very simple with minimal hands-on time. Briefly, the samples are thawed and then centrifuged at 2000xg for 30 minutes to remove cells and debris. The supernatant is then transferred to a new tube and 0.2 volumes of the Exosome Isolation Reagent added. This is incubated at 2-8C for 30 minutes and then centrifuged at 10,000xg for 10 minutes. Finally, the supernatant is removed leaving the exosome pellet which can be further analysed in downstream applications.
