I am using this quantitated T.gondii DNA in my undergraduate thesis research, as part of a PCR protocol that will detect the presence of the parasite in various mammals collected in Western North Carolina. This specific product was chosen because this was quantitated, which will help me to gauge the amount of T.gondii in my specimen. I also was trying to keep a tight budget, and this was definitely the best buy for the amount and price of the product.

In the image, these are the product bands at various concentrations from 1.1X10^4 copies/uL at lane 2 (orig concentration) down to 1.1x10^-1 copies/uL at lane 8.

Protocol Overview

The product was used as a positive control in a series of PCR reactions on heart and brain tissues.

Tips

Would recommend separating the DNA into aliquots so as not to freeze/thaw the DNA more than is necessary.

Results Summary

Produced a gradient of DNA dilutions in PCR reactions and gel electrophoresis, to compare with my samples. The bands came out very thick and bright, even at a 1.1 x 10^2 dilution. I also performed an annealing temperature gradient, with B1 gene primers, from 45C to 57C and all 12 bands came out bright.
