My research focuses on expressing T cell receptors using lentiviral vectors. I use Jurkat cells to test the vectors for TCR expression. I detect antigen-specificity using a HLA-peptide tetramer (Tetramer-APC) and TCR using Anti-TCRa/b-FITC.

Results Summary

The Anti-TCRalpha/beta antibody detects all TCRa/b in an antigen independent manner. While used with tetramer for antigen specificity, the antibody can be used to measure TCR avidity / TCR expression on cell lines. There is a small level of background in Mock-transduced cells, but it could also be from the heterogeneity in Jurkat cell line. However, at high enough TCR expression levels, it won't be a problem.
