Experiments were aimed at detecting apoptotic cells in postnatal mouse brain tissue. Cleaved Caspase 3 is a well-known marker for cells undergoing apoptosis in the caspase-dependent pathway.
Immunofluorescence
Floating brain tissue fixed in 4% PFA and cryosectioned to 50um thickness
1:1000 dilution in blocking agent at 4C overnight
5% Goat Serum; 0.5% Triton X-100; PBS
1:1000 dilution of Goat ant-Rabbit Alexa Fluor IgG in PBS

Fluorescence detected via epifluorescent microscopy
Obtained fluorsecent images that could then be analyzed via stereological software.
