I have used the Qiagen QIAquick PCR Purification kit to successfully purify DNA from PCR products, thus removing primers, nucleotides, polymerases etc. using the QIAquick spin columns in a microcentrifuge. These products were suitable for downstream Sanger sequencing.

Protocol Overview

The protocol is quick and easy to follow and makes use of spin-columns. Briefly, the microcentrification method involves addition of Qiagen supplied buffer to the sample which is then applied to a spin column. The DNA binds to the column while impurities are removed via wash steps using the Qiagen supplied buffer. The purified DNA is then eluted into elution buffer or water.The protocol can be adapted to use with a microcentrifuge or vacuum manifold.

Tips

The initial buffer works at an optimal pH and the kit includes an optional pH indicator which can be added to this buffer.

Results Summary
