This antibody was used along with anti-IL-10R blocking antibodies (Purified anti-human CD210 ) to investigate the effect of IL-10 on T cells in dengue infection (published in PLOS Neglected 2013, Malavige et al). We used this antibody and the receptor blocking antibody to see the effects of complete IL-10 blockade in dengue infection. As earlier we found that IL-10 was high in dengue and was associated with severity and also inversely correlated with dengue specific T cell responses, we wanted to see if the responses recovered with IL-10 blockade.

PBMCs from patients with acute dengue infection
PBMCs were incubated for 1 hour before peptide stimulation in an ELISpot assay or an ICS. The concentration was 5microg/ml at 37 C.
Anti-IL-10 antibody and anti IL-10R antibody

IFNg ELISpot assays and intracellular cytokine secretion assays
The antibody worked excellently in ELISpot assays and in FACS analysis in the ICS assays. The IL-10 was completely blocked and we found that dengue virus specific T cell function was restored.
Ease of use and excellent blocker
This antibody is an excellent antibody to be used along with IL-1OR blocking antibody for complete block of IL-10.
