I needed to identify neuronal cell types in mouse brain for stereological analysis. NeuN is a mature neuronal marker that is localized in the nucleus which allows for ease of cell counting.
Immunofluorescence
50 um thick, floating, postnatal mouse brain fixed in 4% PFA overnight at 4C
1:1000 dilution in blocking agent, overnight incubation at 4C
5% normal goat serum + 0.5% Triton X100
1:1000 dilution of secondary antibody in PBS, overnight incubation at 4C

This procedure results in the nuclear labeling of neurons which can be visualized via fluorescent microscopy.
