Aim: to analyse the role of PIAS1 in glioblastoma cells;Specific goal: to analyze the status of PIAS1 after the treatment of glioblastoma cells with cytokines;Reason: available antibody with appropriate price and (apparently) quality
Western Blot
Human glioblastoma (U251) cells
Dilution; 1:500; Time: overnight; Temp.; 4C; constant rotation
5% nonfat milk, 1hr at RT with constant shaking
anti-Rabbit IgG conjugated with HRP (ThermoScientific); dilution: 1:500; time: 1hr; Temp: RT
N/A
Femto Chemiluminescent reagent from ThermoScientific (method:Chemiluminescence)
Here we shown that PIAS1 is presented in U251 cells and activate after 10 min. treatment of these cells with complex of IL-1beta and IFNgamma. The activation continue until 60 min. Since PIAS1 (protein inhibitor of activated STAT) plays a crucial role in the regulation of STAT pathway which is activated in tumor cells, it is important to futher analyse the role of this protein and according signaling pathway in brain tumor cells.
Good quality, suitable for WB
Good antibody, particularly for Western blot
