The I26R is a large, enclosed, front loading, shaking incubator that allows the user to precisely control both shaking speed and incubation temperature. The machine has a digital thermometer control that allows precise regulation of incubation temperature. Importantly, the i26R model features precise control of temperature close to ambient levels. In our lab, we use this incubator to induce and expand yeast cells that require incubation at 20°C and 30°C, respectively. The large capacity of the instrument (76 X 46 cm aluminum shaker plate) allows us to run several experiments simultaneously. An additional advantage of the instrument is that the incubator can be used without the shaking function being engaged. Furthermore, additional incubator units, if required, can be stacked vertically to save lab space.

EBY100 yeast cells in culture, transfected with DNA to produce recombinant mammalian proteins.

Protocol Overview

EBY100 yeast cells are thawed and electroporated with recombinant DNA in an appropriate yeast display vector. Yeast cells are incubated, shaking in the I26R incubator at 30°C for one hour and plated out on selective agar plates for 48h (without shaking), again at 30°C. Individual colonies are selected and transferred to an induction medium and incubated at 20°C for 24h. The cells are washed and then re-suspended in an expansion media for an additional 30°C for 24h. Yeast cells are then analyzed, sorted and those expressing the protein of interest are selected and analyzed.

Tips

The adjustability of the digital thermometer and shaking control on this incubator make it easy to adjust the temperature, even in a relatively short time (<10 mins). We have found that placement of the incubator on a lab bench is ideal although it’s footprint will require a deep bench space. If more than one incubator is planned, the first would be best placed on the floor. One drawback that we have encountered is that access to the front of the incubator can be difficult when the door is down (it opens to the front). Shorter members of the lab sometimes struggle to place large or heavy flasks at the back of the incubator.
