New research from the Del Monte Institute for Neuroscience at the University of Rochester has uncovered unexpected sex-related differences in microglia function in adult mice. This finding challenges the previous assumption that microglia behave similarly across sexes in adulthood and could have significant implications for the study and treatment of neurodegenerative diseases like Alzheimer's and Parkinson's.
The study, published in Cell Reports, focused on the response of microglia to an enzyme inhibitor, PLX3397, which blocks the microglia survival receptor. Researchers observed that while male mice exhibited the expected depletion of microglia when exposed to PLX3397, female mice showed a different signaling strategy resulting in increased microglial survival and less depletion.
Ania Majewska, senior author of the study, commented, "It is a fortuitous finding that has repercussions for what people are doing in the field, but also helps us understand microglia biology in a way that people may not have been expecting."

This discovery highlights the importance of gender-specific research in neuroscience, particularly given the known disparities in Alzheimer's and Parkinson's diagnoses between men and women.
“These findings are crucial in the rapidly emerging field of developing disease-modifying therapies that target microglia,” said Majewska. “We do not yet know why the microglia are acting differently in the two sexes. I think we'd like to understand how the signaling through this receptor is regulated in different conditions, i.e. hormonal changes, basal state, inflammatory, or an anti-inflammatory state.”
