New research from RCSI University of Medicine and Health Sciences has found an intricate relationship between the body's internal clock and the immune system's inflammatory process. The study, which focused on macrophages, revealed that the activation of the NLRP3 inflammasome, a complex within macrophages that triggers inflammation, is not constant throughout the day but is regulated by the body's circadian rhythms. This 24-hour cycle influences when macrophages are most efficient at detecting threats and when their energy levels peak for mounting a response.
According to Annie Curtis, senior author of the paper published in The FASEB Journal, "When macrophages 'think' it's morning, their inflammasome activation is quicker and more robust." This heightened immune response during the early part of the day coincides with when humans are more likely to encounter environmental challenges such as injuries or infections.

The study also highlights the significant role of mitochondria, the cell's energy producers, in driving these daily changes in immune activity. This finding provides new insights into why symptoms of inflammatory diseases like arthritis often worsen in the morning.
James O'Siorain, the study's lead author, suggests that these findings could lead to more refined treatments for inflammatory conditions. By targeting inflammasomes at specific times of the day when macrophage activity peaks, new therapies could potentially be more effective.
