New research from ETH Zurich has uncovered the molecular basis for the frustrating yo-yo effect experienced by many dieters. The study, published in Nature, reveals that fat cells retain an epigenetic memory of obesity, making it easier for individuals to regain weight after losing it.
Epigenetics plays a crucial role in determining gene activity in cells. Unlike genetic sequences, epigenetic markers can be influenced by environmental factors and eating habits. The researchers found that obesity leads to characteristic epigenetic changes in fat cell nuclei that persist even after weight loss.
"The fat cells remember the overweight state and can return to this state more easily," explains Ferdinand von Meyenn, senior author on the study. This memory effect was demonstrated in mice, which regained weight more quickly when re-exposed to a high-fat diet after initial weight loss.

The team also found evidence of this mechanism in humans by analyzing fat tissue biopsies from formerly overweight individuals who had undergone weight loss surgery. While they examined gene expression rather than epigenetic markers in human samples, the results were consistent with the findings in mice.
Currently, there is no way to erase this epigenetic memory with drugs. Laura Hinte, first author of the study, notes that fat cells live for an average of ten years before being replaced. Given this long-lasting effect, the researchers emphasize the importance of preventing obesity in the first place, especially in children and young people.
The study opens up new avenues for research, as the team suspects that other body cells might also contribute to the yo-yo effect. Future investigations will explore whether cells in the brain, blood vessels, or other organs similarly remember obesity and influence weight regain.
