ELISA stop solutions are ready-to-use reagents used in ELISA applications in order to inactivate the detection enzyme of the immunoassay. The stop solution is added to each well of the ELISA plate, causing a drastic shift in the pH and terminating the ongoing reaction. This stabilizes the color change of the medium, allowing a period in which the optical density of the assay can be accurately measured. The stop solution should not affect the background or optical density readings. Stop solutions are a safer substitute for the traditional acids that have been commonly used for this purpose.
