Western blot was performed using Anti-CDK2 Monoclonal Antibody (Product # MA5-32017) and a 34 kDa band corresponding to CDK2 was increased upon Hydroxyurea treatment in HCT-116 cell line and was also observed in Mouse Thymus tissue extracts but not in Mouse Liver as reported in literatures. Whole cell extracts (30 µg lysate) of HCT-116 (untreated) (Lane 1), HCT-116 treated with Hydroxyurea (2mM for 20 hours) (Lane 2), tissue extracts of Mouse Thymus (Lane 3) and Mouse Liver (Lane 4) were electrophoresed using Novex® NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:2000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
