Immunohistochemical analysis of IL-29 in paraffin-embedded rat brain. Sample was incubated with IL-29 polyclonal antibody (Product # PA5-87056) using a 1:100 dilution. Images are presented at 40x magnification.

Western blot was performed using Anti-IL-29 Polyclonal Antibody (Product # PA5-87056) and a 22 kDa band corresponding to Interferon lambda-1 was in the treatment panel tested. No band was observed in U-937 cells treated with protein transport inhibitor which is a negative expresser of IL-29. Whole cell extracts (30 µg lysate) of THP-1 (Lane 1), THP-1 differentiated to M2 macrophages treated with Human IFN Alpha, Lipopolysaccharide and Protein transport inhibitor (Lane 2), THP-1 differentiated to M2 macrophages treated with Human IFN Alpha, Polyinosinic–polycytidylic acid sodium salt and Protein transport inhibitor (Lane 3), THP-1 treated with Protein transport inhibitor (Lane 4), U-937 (Lane 5) and U-937 treated with Protein transport inhibitor (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:20,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescentdetection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). An uncharacterized band (*) at ~70 kDa was also observed in a few cell lines.
