Western blot analysis was performed on whole cell extracts (30 µg lysate) of A549 (Lane 1), A-431 (Lane 2), HeLa (Lane 3) Hep G2 (Lane 4) and tissue extracts (30 µg lysate) of Mouse Heart (Lane 5) and Mouse Brain (Lane 6). The blot was probed with Anti-MST2 Recombinant Rabbit Monoclonal Antibody (Product # 703027, 1:5000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.4 µg/mL, 1:4000 dilution). A 59 kDa band corresponding to MST2 was observed across the cell lines and tissues tested.
Knockdown of MST2 was achieved by transfecting A549 cells with MST2 specific validated siRNA (Silencer® select Product # s13568). Western blot analysis was performed using membrane enriched extracts from the MST2 knock down cells (Lane 3), non-specific scrambled siRNA transfected cells (Lane 2) and untransfected cells (Lane 1). The blot was probed with Anti-MST2 Recombinant Rabbit Monoclonal Antibody (Product # 703027, 1:5000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.4 µg/mL, 1:2500 dilution). Loss of signal upon siRNA mediated knock down confirms that antibody is specific to MST2.
