Description
FAD4T was generated by introducing human APP carrying the Swedish and Indiana mutations together with human PSEN1 carrying the M146L and L286V mutations into the mouse genome. The model develops rapid-onset Alzheimer's disease pathology, with Aβ deposition detectable at 2–3 months of age and pathology comparable to 5XFAD mice by 4 months, accompanied by neuroinflammation and synaptic loss. It is suitable for studies of Alzheimer's disease pathogenesis and long-term pharmacodynamic evaluation and has been validated in studies including aducanumab. A single-parent breeding strategy (♂FAD4T × ♀WT) provides stable and consistent pathology in the offspring
