Assay Principle
Isothermal amplification kits enable exponential replication of a target nucleic acid sequence at a single, constant temperature, removing the need for the thermal cycling equipment required by conventional PCR. Loop-mediated isothermal amplification (LAMP), the most widely commercialized isothermal amplification assay format, relies on a strand-displacing DNA polymerase together with a set of four to six specially designed primers that recognize six to eight distinct regions of the target sequence, generating characteristic stem-loop DNA structures that drive rapid, exponential amplification at a fixed temperature, typically around 60 to 65°C. RT-LAMP master mixes incorporate a reverse transcriptase alongside the strand-displacing polymerase in a single reaction, allowing direct amplification from RNA templates without a separate cDNA synthesis step. Detection is commonly achieved through one of two readouts: a colorimetric format that exploits the pH shift produced by amplification (typically visualized through a phenol red or similar pH-sensitive indicator that shifts from pink to yellow), or a fluorescent format using an intercalating or sequence-specific probe-based dye that permits real-time monitoring on a fluorescence-capable instrument.
Protocol highlights and purchasing considerations
Protocol Highlights
LAMP has become a mainstay of rapid pathogen detection, point-of-care diagnostics, and field-deployable testing, and RT-LAMP variants extend this capability to RNA targets such as viral genomes. Reaction times are typically fast, often producing a visible or measurable signal within 15 to 60 minutes, and because amplification occurs at a single temperature, these assays can run on simple heat blocks or water baths rather than requiring a thermal cycler, an advantage for field or point-of-care settings with limited infrastructure. Colorimetric master mixes generally require carefully buffered, low-bicarbonate sample input, since residual buffering capacity from certain sample types or extraction methods can mute or obscure the pH-based color shift; some kit formats include a UDG (uracil-DNA glycosylase) component to reduce carryover contamination risk between reactions. Primer design is considerably more involved than for standard PCR, so most researchers rely on pre-validated, target-specific primer sets from a kit supplier or established design tools rather than designing LAMP primers from scratch.
Assay Kit Purchasing Considerations
Colorimetric and fluorescent detection formats used in isothermal amplification kits represent the primary choice point, with colorimetric formats favored for visual, equipment-light workflows and fluorescent formats favored where quantitative or real-time kinetic data is needed. Because LAMP is prone to false-positive signal from primer-dimer or non-specific amplification if reaction conditions are not tightly controlled, researchers should confirm that a kit's validated primer sets and reaction protocol have been tested against the specific target sequence and sample matrix of interest, rather than assuming general LAMP master mix performance will translate directly. For RNA viral targets, confirming that an RT-LAMP master mix has been validated for direct use with crude or minimally processed sample types (saliva, nasal swab eluate) is worth checking if the intended workflow aims to skip a separate RNA extraction step, since not all master mixes tolerate the same degree of sample complexity. As with any activity assay, it's good practice to review the full protocol before deciding on a kit, since the manual often reveals practical details, such as incubation times, control requirements, and data analysis steps, that aren't captured in the product summary alone.
