Inorganic phosphorus (Pi) is a fundamental component of nucleic acids, phospholipids, and high-energy compounds such as ATP, and circulating phosphate levels are routinely monitored in renal, bone, and metabolic research. A phosphorus assay typically relies on the classic phosphomolybdate reaction, in which inorganic phosphate reacts with molybdic acid to form phosphomolybdic acid, which is then reduced to a blue-colored molybdenum blue complex under the action of a reducing agent, with absorbance read around 660 nm and intensity proportional to phosphorus concentration. A protein precipitation step is typically included ahead of the color reaction to remove serum or plasma proteins that could otherwise interfere with the readout.
