Matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases that degrade extracellular matrix components including collagen, gelatin, elastin, and fibronectin. An MMP assay generally follows one of two general approaches depending on the research question. Universal MMP activity kits use a broadly cleavable fluorogenic substrate that most family members can process after activation with an organomercurial compound such as APMA, providing a general readout of total MMP-like proteolytic activity in a sample, with fluorescence typically read around Ex/Em 490/525 nm. Isoform-specific kits instead use a FRET-based peptide substrate designed to be preferentially cleaved by a single MMP (such as MMP-3), releasing a quenched fluorophore for detection at isoform-specific excitation and emission wavelengths (for example, Ex/Em 325/393 nm for an Mca-based MMP-3 substrate). Because most MMPs have overlapping substrate specificity, achieving true isoform selectivity generally requires either a purified recombinant enzyme system or a substrate carefully validated against cross-reactivity with related family members.

Universal activity kits are commonly used for MMP inhibitor screening when paired with a purified MMP enzyme preparation, since the assay format allows straightforward IC50 determination from a dose-response curve. Because endogenous tissue inhibitors of metalloproteinases (TIMPs) are present in most biological samples and can suppress measured activity, some protocols recommend an activation step with APMA or a similar compound to fully unmask latent (pro-) MMP activity before assay readout.

The primary choice when selecting an MMP assay kit is universal versus isoform-specific detection, and this should be driven directly by whether the research question requires total MMP-like activity or activity attributable to a single family member. For cancer or arthritis research focused on a specific MMP, confirming that a kit's substrate has documented selectivity data (rather than simply being marketed for that isoform) is worth checking, given the substrate promiscuity common across this enzyme family. Researchers running inhibitor screening campaigns should also confirm that a kit's fluorogenic substrate and reaction buffer are compatible with the specific recombinant MMP construct and any solvent carriers used to deliver test compounds, since DMSO and other common vehicle solvents can interfere with certain fluorescence-based readouts above a threshold concentration. As with any activity assay, it's good practice to review the full protocol before deciding on a kit, since the manual often reveals practical details, such as incubation times, control requirements, and data analysis steps, that aren't captured in the product summary alone.
