Gamma-glutamyl transferase (GGT) catalyzes the transfer of gamma-glutamyl groups between peptides and amino acids, playing a central role in glutathione metabolism and amino acid transport across cell membranes. A GGT assay typically uses a synthetic gamma-glutamyl donor substrate (glutamyl p-nitroaniline) together with an acceptor (typically glycylglycine); GGT catalyzes transfer of the gamma-glutamyl group to the acceptor, releasing free p-nitroaniline, a chromogenic product with a characteristic absorption peak at approximately 405 nm. Activity is calculated from the rate of absorbance change over time, providing a direct kinetic readout of enzymatic turnover without requiring a secondary coupling reaction.
