Choline acetyltransferase (ChAT) catalyzes the biosynthesis of acetylcholine from acetyl-CoA and choline, serving as the rate-limiting enzyme for cholinergic neurotransmission. A ChAT assay typically uses a coupled colorimetric reaction rather than direct product detection to measure this activity. As ChAT catalyzes the transfer of an acetyl group from acetyl-CoA to choline, it generates coenzyme A (CoA) as a byproduct, and this free CoA reacts with 4,4'-dithiopyridine to produce a chromogenic signal read at approximately 324 nm.
