Alpha-L-fucosidase (AFU, encoded by FUCA1) is a lysosomal hydrolase that removes terminal alpha-L-fucose residues from glycoproteins, glycolipids, and oligosaccharides. An AFU assay generally uses one of two synthetic substrate formats: colorimetric kits use a nitrophenyl-alpha-L-fucoside substrate, which AFU hydrolyzes to release yellow nitrophenol with peak absorbance around 405 nm, read on a standard microplate reader.
