Parallel Artificial Membrane Permeability Assay (PAMPA) kits are available from a number of suppliers for the testing of drug permeability. A PAMPA kit generally features two aqueous buffer solution holes separated by an artificial membrane. Some membranes imitate the gastrointestinal mucosa and are built of 3 layers. The upper and lower layers are regenerated cellulose hydrates, which cover the middle lipid layer. The test compound is diluted in the buffer and placed in the donor well, where it will cross the membrane through passive diffusion, and exit from the acceptor hole. The effective permeability of the compound is used to determine the rate of permeation. Testing drug permeability by cellular methods can be expensive and time-consuming, therefore the use of PAMPA kits circumvents these issues. PAMPA kits are used in particular to test the gastrointestinal membrane permeability of potential clinical compounds.
