
ELISA blocking buffers are solutions designed to passively adsorb to all remaining binding surfaces of the assay plate that do not contain a coated protein. Blocking buffers remove the possibility of non-specific binding and therefore help to increase stability, reduce background noise and make the assay more sensitive. ELISA blocking buffers should not interfere with the assay itself, and should only contain compounds that do not themselves react with the epitope. There are multiple types of ELISA blocking buffers available, each with its own advantages and disadvantages. Protein blockers are a permanent blocking solution, while detergent-based buffers are temporary and can be removed by washing. Non-protein blocking buffers offer advantages such as minimizing the cross-reactivity that may occur with protein buffers. Single protein buffers also show lower cross-reactivity when compared to serum solutions or combination buffers. Some ELISA buffers can act as diluents for the assay as well.
