
Buffers for use in SDS-PAGE are available from several suppliers. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) is an analytical technique used to separate proteins based on their molecular weight. Loading buffers are important when preparing samples to be loaded into the gel for electrophoresis. Loading buffers can vary in composition but often contain Tris-HCL, SDS, Bromophenol blue, and glycerol. SDS denatures proteins and adds a net negative charge for movement in an electric field. Bromophenol blue is used to visualize the run of the proteins on the gel. Glycerol increases the density of the sample to make loading easier and Tris helps to maintain the pH. ß-mercaptoethanol or dithiothreitol (DTT) can also be added to the buffer to break disulfide bonds and maintain the unfolded state.
